Aging is the most complex of phenotypes that occur in humans and is associated with a plethora of pathologies. Recent studies have suggested that there are specific alterations in metabolism shared between aging organisms and cancer cells. Increased lactate production with aerobic glycolysis is seen in aging and is a hallmark feature of cancer cells, a metabolic phenomenon called the Warburg effect. Importantly, this effect is also seen after DNA damage and during hypoxia.  One possible factor involved in the development of the Warburg effect is the transcription factor HIF-1alpha, a factor involved in the cellular adaptation to hypoxia.

 

The goal of my project is to understand the interplay between hypoxia, HIF-1alpha and DNA damage and how this interplay may be regulated in the aging process. Artificial intelligence algorithms, classic molecular biology techniques, metabolomics and proteomics approaches will be utilized. Further, in order to translate these discoveries into humans, I will conduct population studies involving Ethiopian high- land dwellers and low- land dwellers.  Combined, my project will provide evidence of the underlying processes that lead to aging and age-associated diseases.
